If you are using Virtuoso Layout Editor, first you have to select the PRBoundary(Usually Blue in Color but depends) layer in the Layer Selection Window(LSW) and draw a rectangle as per your need.

Are you using IC5 or IC6 ? In IC6 you have to use Menu 'Create->P&R Object->P&R Boundary'. As said by achitrag in IC5 you have to select the prBoundary layer in LSW windows and then draw a rectangle. If the layer is not listed you may need to enable it by using the menu in the LSW window 'Edit->Set Valid Layers...'
The PR boundary is the layer that the place and route tools will use for placing the cells next to each other. So if you have overlapping ground and vdd lanes it can be that you have the contacts put in such a way that they half above and half below the PR boundary.
